Being very efficient catalysts, enzymes are essential for all biological processes and find a broad range of applications in the chemical and pharmaceutical industry. Despite their importance, still little is known about how enzymes evolve. In recent years a large number of enzymes with so-called promiscuous activities have been discovered. These enzymes are able to catalyze other reactions in addition to their natural activity. It might be considered that this promiscuous activity serves as a starting point for the evolution of a new catalytic activity.

You will analyze promiscuous enzymes with a number of different approaches to study the evolvability of the promiscuous activity and to understand its structural origin. The project involves the expression and purification of enzymes and eventually the synthesis of enzyme substrates. You will further develop bioconjugation methods to immobilize the enzymes to surfaces and develop novel enzyme activity assays. The ultimate goal is the characterization of the enzymes with single molecule fluorescence experiments using substrates for both the natural and the promiscuous activity.

We are searching for highly motivated candidates with a strong background in protein chemistry, protein engineering/design, molecular biology or chemical biology. Experience in more than one of these areas is highly appreciated. The project is multidisciplinary and involves collaborations with other groups at the institute as well as a number of external collaborations. The candidates should have a strong interest in a multidisciplinary, collaborative approach. Team spirit and communication are crucial for a successful development of the projects.
